I tried to test run the Desktop Media Uploader after hearing so many positice comments, I don't have a live site to test it against so I tried hitting localhost, the timer just spins and spins and nothing happens on login, where am I going wrong?
Could you install something like fiddler (http://www.fiddler2.com/fiddler2/) to see what request is being made to the server and what the response it gets is?

The package should work fine on localhost.

ps It can sometimes take a while if the app pool has recently been recycled.
